Abstract
This paper details the development and use of a reverse phase high performance liquid chromatographic (RP-HPLC) technique for the am/no acid analysis of proteinaceous artists' media. Samples (for example, animal glues, milk and egg proteins) were hydrolysed under acid conditions, then derivatized with 9-fluorenyl-methyl chloroformate (FMOC). Analysis was achieved via injection onto a Spherisorb ODS-2 (Phase Separations Ltd, Deeside, Flintshire, UK) column with a gradient elution programme and ultraviolet (UV) detection. Samples of proteinaceous binding materials removed from works of art have been successfully identified using the method described.
